Cleaning workers vulnerable to work fatigue due to the high physical demands of their daily tasks. This high physical workload has the potential to lead to job fatigue. Job fatigue can reduce productivity, increase the risk of workplace accidents, and had negative impact effect on overall worker health. Age and physical workload to have a significant relationship with the incidence of job fatigue. This study aimed to determine the relationship between age and physical workload level and work fatigue among cleaning workers. This study used a quantitative method with a cross-sectional approach involving all 39, chosen using a total sampling technique. The research tool used is the Omron digital device, Fatigue Assessment Scale (FAS) questionnaire, and data were analyzed using the Chi-Square test. The statistical tests showed that there was no relationship between age (p-value = 0.234) and work fatigue, and a relationship between physical workload and work fatigue (p-value = 0.008). This study concluded that there was no significant relationship between age and work fatigue, and there was a significant relationship between physical workload and work fatigue among cleaning staff at the Grhasia Mental Hospital, Yogyakarta
